What is Reta?

Reta is a synthetic 39-amino-acid peptide. It is the first triple agonist to be studied in large clinical trials: a single molecule that activates three different hormone receptors. That makes it the successor to the single GLP-1 agonists and to the dual agonist Tirze.

A fatty acid tail (a C20 diacid) is attached to the peptide chain. It binds to albumin in the blood, so the peptide is broken down and cleared from the body only slowly. Its half-life is around six days, which is why Reta was given once a week in the clinical trials.

How Reta works: three receptors at once

Reta’s mechanism centres on three hormones the body produces itself around meals and when energy is in short supply. Reta mimics all three:

  • GLP-1 receptor. GLP-1 (glucagon-like peptide-1) is released by the gut after eating. It boosts insulin release when blood sugar rises, slows gastric emptying and sends a satiety signal via the brain.
  • GIP receptor. GIP (glucose-dependent insulinotropic polypeptide) also boosts insulin release and is involved in how fat is stored and processed in adipose tissue.
  • Glucagon receptor. Glucagon prompts the liver to release energy. In research, activation of this receptor has been linked to higher energy expenditure and to the breakdown of fat in the liver.

At the GIP receptor, Reta is more potent than the body’s own GIP, while at the GLP-1 and glucagon receptors it is weaker than the natural hormones. That balance is deliberate: enough glucagon activity for extra energy expenditure, without throwing blood sugar off course.

What does the research on Reta show?

Reta is one of the best-documented new peptides. The key published studies:

  • Phase 2 in obesity (NEJM, 2023). 338 adults with obesity or overweight received Reta or placebo for 48 weeks. In the highest dose group, body weight was on average 24.2% lower, compared with 2.1% in the placebo group. At the time, this was the largest effect measured in a phase 2 trial of an obesity drug.
  • Phase 2 in type 2 diabetes (The Lancet, 2023). In 281 participants, the long-term blood sugar marker (HbA1c) fell by more than 2 percentage points after 36 weeks in the highest dose groups.
  • Fatty liver (Nature Medicine, 2024). In a substudy of people with high levels of liver fat, liver fat had returned to a normal level (below 5%) in most participants in the higher dose groups after 48 weeks.
  • Phase 3 (TRIUMPH programme). Since 2023, a series of large phase 3 trials has been under way in areas including obesity, type 2 diabetes, knee osteoarthritis and sleep apnoea. Reta is not registered as a medicine in Europe.

The most frequently reported side effects in these studies were gastrointestinal complaints such as nausea, diarrhoea and vomiting, mainly during the period when the dose was being built up. A dose-dependent increase in heart rate was also observed.

Which Reta vial should you choose?

All vials come from the same production batch; only the amount of peptide per vial differs. The larger the vial, the lower the price per milligram, so the 30 mg vial offers the best value per mg. A smaller vial is handy if you are planning a shorter experiment or want to compare different batches. You can work out the concentration after dissolving with the formula.

Quality you can check for yourself

With a research peptide, purity determines whether your results can be trusted. That is why every batch of Reta goes through the same checks before it goes on sale:

  • Identity by mass spectrometry: is there really Reta in the vial, with the correct molecular weight?
  • Purity by RP-HPLC: this batch reaches 99.4%. We do not sell batches below 99%.
  • Moisture, endotoxins and residual solvents are measured for every batch.

All results are in the COA on the product page. To learn how to read one, see Reading a Certificate of Analysis in five steps. And what that 99% actually tells you is explained in What does 99% purity really mean for peptides?

Reconstituting and storing Reta

Reta is supplied lyophilised (freeze-dried), because that is when the peptide is most stable. For use in the lab, dissolve the powder in bacteriostatic water: add it slowly down the side of the vial and swirl gently, don’t shake. The steps are set out in Reconstitution: from lyophilizate to solution.

  • Lyophilised: keeps for 24 months at 2–8 °C, dark and dry.
  • After reconstitution: store in the fridge, protected from light, and use within a few weeks.
  • Avoid repeated freezing and thawing.

For more background, see Storage and stability of lyophilized peptides.

Tirze activates two receptors: GLP-1 and GIP. Reta also activates the glucagon receptor, which makes it a triple agonist. In research, that extra glucagon activity has been linked to higher energy expenditure and to the breakdown of liver fat.

Reta mimics three of the body's own hormones: GLP-1, GIP and glucagon. GLP-1 and GIP boost insulin release and play a role in satiety and fat storage; glucagon prompts the liver to release energy. A fatty acid tail on the molecule gives it a half-life of around six days.

The 30 mg vial has the lowest price per milligram. All vials come from the same batch, so the only choice you are making is the amount.

Add the bacteriostatic water slowly down the side of the vial so the powder doesn't foam. Then swirl gently until everything has dissolved; don't shake.

Lyophilised Reta keeps for 24 months at 2–8 °C, dark and dry. After reconstitution, store the solution in the fridge, protected from light, and use it within a few weeks.

If the product is in stock and you order on a working day before 16:00, we ship it the same day with PostNL.
